Soluble Relaxing Factor from Muscle*
نویسنده
چکیده
Gergely, Briggs, and Kaldor (24) have recently shown that the so called Marsh-Bendall relaxing factor consists of a granular component and a heat labile dialyzable cofactor. The requirement for both components could be demonstrated for the inhibition of Mg++-activated myofibrillar adenosine triphosphatase, for the inhibition of the adenosine triphosphate-induced syneresis of myofibrils and for the reversal of the ATP-induced tension development of glycerol extracted muscle fibers. This report is concerned with the description of a new granulefree system obtained after incubation of granules and cofactor in the presence of ATP which duplicates the action of the complete relaxing factor system (RFS)f A number of properties of the soluble relaxing substance (RS) will be discussed.

It has been reported by several groups of workers that incubation of the relaxing factor granar of muscle with magnesium and adenosine triphosphate results in the formation of a soluble relaxing substance capable of inhibiting the magnesium-activated adenosine triphosphatase activity of myofibrils or actomyosin (3-6) and the contraction of single glycerinated muscle fibers (7). In addition, Uri...

Endothelium-derived relaxing factor and exogenous nitrovasodilators are thought to produce smooth muscle relaxation by activation of soluble guanylate cyclase. To investigate whether diminished cyclic GMP (cGMP) accumulation underlies the differences in vascular reactivity to nitrovasodilators between Wistar-Kyoto (WKY) rats and spontaneously hypertensive rats (SHR), we determined cGMP formatio...
متن کاملThe Role of Calcium in the Adenosine Triphosphatase Activity of Myofibrils and in the Mechanism of the Relaxing Factor System of Muscle*
Several factors tend to create the impression that the removal of Ca from the myofibrils plays an important role in the mechanism of relaxation. Among these factors are the inhibition of the activity of the relaxing factor system of muscle by Ca (2) ; the similarity between the effect of ethylenediaminetetraacetate and the relaxing factor system in producing inhibition of myofibrillar adenosine...
متن کاملStimulation of soluble guanylate cyclase by an acetylcholine-induced endothelium-derived factor from rabbit and canine arteries.
The present study was designed to investigate the hypothesis that, during acetylcholine-induced endothelium-dependent relaxation, a factor(s) is released from endothelial cells which directly activates soluble guanylate cyclase. We attempted to determine what similarities or differences existed between this factor and endothelium-derived relaxing factor. The study was performed on segments of r...
